ABSTRACT

The aim of this study was to determine the effects of extremely low frequency electromagnetic field (ELF-EMF) on energy metabolism and oxidative stress in Caenorhabditis elegans (C. elegans). Worms in three adult stages (young adult stage, egg-laying stage and peak egg-laying stage) were investigated under 50 Hz, 3 mT ELF-EMF exposure. ATP levels, ATP synthase activity in vivo, reactive oxygen species (ROS) content, and changes of total antioxidant capacity (TAC) were detected, and worms' oxidative stress responses were also evaluated under ELF-EMF exposure. The results showed that ATP levels were significantly increased under this ELF-EMF exposure, and mitochondrial ATP synthase activity was upregulated simultaneously. In young adult stage, worms' ROS level was significantly elevated, together with upregulated TAC but with a decreased ROS-TAC score indicated by principal component analysis. ROS level and TAC of worms had no significant changes in egg-laying and peak egg-laying stages. Based on these results, we concluded that ELF-EMF can enhance worm energy metabolism and elicit oxidative stress, mainly manifesting as ATP and ROS level elevation together with ATP synthase upregulation and ROS-TAC score decrease in young adult C. elegans.

ABSTRACT

Leiomyosarcoma(LMS)of the great saphenous vein(GSV)is a rare condition that is often misdiagnosed due to the lack of typical clinical manifestations.This article reported a GSV-LMS case diagnosed and treated in Peking Union Medical College Hospital.Meta-analysis of the current case and 41 cases derived from CNKI,WANFANG DATA,and MEDLINE database was also conducted,which revealed that the male to female ratio of GSV-LMS was 11:10;the median age of disease onset was 59.5 years;the most commonly affected region was thigh.Due to lack of specific clinical features,42.9% of GSV-LMS patients were misdiagnosed at presentation.Radical resection is the most effective therapy,and the indications,effectiveness and protocols of adjuvant radiochemotherapy remain unclear.The 3-and 5-year overall survival rate was 86.1% and 77.5%,respectively.Recurrence and metastasis occurred in 31.0% of patients after surgery.

ABSTRACT

Objective To investigate the function of autophagy in the process of PM2.5-induced apoptosis. Methods PM2.5 was obtained from Zhanjiang in 2016. Human lung adenocarcinoma cells H441 were treated with PM2.5 at different concentrations for different times. Cell proliferation was analyzed by MTT assay; Cell apoptosis was assessed by PI and Annexin V double staining and TUNEL assay. The expression of autophagy marker LC3Ⅱ, AKT and P-AKT protein was examined by Western blot ( WB). H441 cells were treated with PM2.5 following treatment with rapamycin or 3-MA. Cell viability was evaluated by trypan blue staining. Results Compared with the control group, cell proliferation was significantly inhibited by PM2.5 at concentration of 100 μg/mL or more for 24 and 48 h. With the increase of PM2.5 concentration, the cells apoptotic rate significantly increased, the protein ex-pression of LC3Ⅱwas increased as well as the P-AKT was decreased; and the protein expression of LC3Ⅱwas in-creased significantly after AKT inhibitor treatment. Moreover, rapamycin decreased PM2.5-induced cell apoptosis, and 3-MA can promote PM2.5-induced cell apoptosis. Conclusions In H441 cells, PM2.5 activates autophagy by inhibiting activation of AKT pathway, and cell autophagy can mitigate PM2.5-induced apoptosis.

ABSTRACT

There is high demand for a sensitive method for miRNA detection in clinical diagnosis. In this work, we developed a method for miRNA detection based on the surface plasmon-enhanced energy transfer ( SPEET ) between gold nanoparticles ( AuNPs ) and silver nanoclusters ( AgNCs ) , coupled with DNA polymerase and nicking enzyme-assisted isothermal amplification for target recycling. Two DNA probes ( Probe a and Probe b) were assembled onto the surface of AuNPs to form Probe b-Probe a-AuNP conjugates. Probe a consisted three domains:the complementary sequence of miRNA, the specific site of the nicking enzyme, and the self-assembly sequence for AgNCs. The 3′ end of Probe a was modified with thiol as a binding site for AuNPs. The SPEET of AgNCs and AuNPs was inhibited when miRNA was added to produce the dumbbell shaped template by polymerase. The template could promote synthesis of AgNCs, resulting in replacement and subsequently recycling of the target molecule for signal amplification. In comparison with the traditional method of miRNA detection with commercial RT-PCR kits, this method avoided the process of reverse transcription and was easy to perform. In addition, this method with a detection limit of 2. 5×10-11 mol/L was cost-effective, label-free, and highly selective for detecting miRNA, and could be applied to the analysis of miRNA in biological samples.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the sonographic characteristics of intraveous leiomyomatosis (IVL) with intracardiac extension and improve its diagnosis.</p><p><b>METHODS</b>The clinical and sonographic data of 13 female patients with pathologically confirmed IVL with intracardiac extension who were treated in our hospital between 2002 and 2012 were retrospectively analyzed. These patients aged 44 years old (range: 38-49 years), and 10 of them were first-episode patients and the remaining 3 were recurrent patients. Eight patients had a history of hysterectomy for leiomyoma.</p><p><b>RESULTS</b>The first-episode symptoms included exertional chest tightness and shortness of breath (n=5), abdominal distention and edema of low extermity (n=4), exertional palpitation of cardiac origin (n=3), and menorrhagia (n=1). Ultrasonography showed that all patients had isoechoic or hypoechoic tumors extended through the inferior vena cava into right heart chambers (62% in right atrium alone and 38% in right ventricle and atrium). Nine masses in right heart chamber (69.2%) were oval and 4 (30.8%) were serpentine, which were all with well-demarcated borders and most (80%) with heteroechogenic texture. Ten patients had hypoechoic or mixed echoic tumors in pelvic cavity or uterus, and 6 of them had abundant blood flow.</p><p><b>CONCLUSIONS</b>IVL with intracardiac extension has certain sonographic characteristics. Ultrasonography is a valuable tool in the diagnosis of IVL with intracardiac extension.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To summarize the sonographic features of the lymphoma in the abdominal lymph nodes.</p><p><b>METHOD</b>The clinical data and sonographic findings of 41 lymphoma patients with original appearance of abdominal mass or lymphadenectasis were retrospectively analyzed.</p><p><b>RESULTS</b>Among these 41 cases, the retroperitoneal and mesenteric lymph nodes were most commonly involved.These patients were divided into two types according to sonographic appearances:solitary mass(n=14) and multiple nodules(n=27) .Solitary hypoechoic mass in abdomen was found in the former type, mostly with irregular or lobular shape, and the inner echo was often heterogenous.The typical appearances of the latter type were multiple enlarged lymph nodes, mostly round or oval, with homogeneous inner echo and clear margins.Some other sonographic characteristics were also helpful for the diagnosis of lymphoma, such as cobblestone sign, intranodular reticulation, vessels-embedded sign, and hilar vascularity.</p><p><b>CONCLUSION</b>Ultrasonography can provide useful information in the diagnosis of the lymphoma in the abdominal lymph nodes.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the distribution of monocyte subpopulations and the changes of Toll-like receptor 4 (TLR4), CD163 and CD36 expressions in the peripheral blood of patients with hyperlipidemia of phlegm-stagnancy obstruction syndrome pattern (HLE-PSO), and to evaluate the intervening effects of Qutan Huayu Jiedu (QHJ) herbs on these parameters.</p><p><b>METHODS</b>Monocytes in the peripheral blood were sorted using flow cytometry into 3 subpopulations: the CD14high CD16- (Mo1), the CD14high CD16+ (Mo2a) and the CD14low CD16+ (Mo2b) subpopulation. The percentages of various monocyte subpopulations in 83 patients and 42 matched healthy controls were determined and the levels of their surface receptors TLR4, CD163 and CD36 expressions were assayed with flow cytometer. Furthermore, patients allocated in the tested group (10 patients) and the control group (11 patients) were treated with QHJ Herbs and Qutan Huayu (QH, removing Phlegm and dissolving stagnancy but without detoxication) herbs respectively. The changes of monocyte subpopulations percentage and TLR4, CD163 and CD36 expressions were determined 4 weeks after they received treatment.</p><p><b>RESULTS</b>Percentage of Mo2a subpopulation was significantly higher in HLE-PSO patients than the normal range (5.35 +/- 2.57 vs. 3.09 +/- 2.38, P < 0.01), but the deviation of the other two subpopulations in percentage was insignificant. The TLR4 expression on Mo1 monocyte in patients was lower than normal (50.73 +/- 24.45 vs. 69.92 +/- 21.06, P < 0.01), while CD163 and CD36 expressions of all three subpopulations in patients were similar to those in healthy persons respectively. After 4 weeks of treatment, a significant lowered Mo2a proportions were observed in the tested group (3.73 +/- 1.05 vs. 5.50 +/- 2.06, P = 0.043); but not in the control group (4.20 +/- 1.81 vs. 5.65 +/- 1.89, P = 0.097), while the levels of TLR4, CD163 and CD36 were not significantly altered after treatment in both groups.</p><p><b>CONCLUSIONS</b>The elevated proportions of Mo2a subpopulation and the lowered TLR4 expression in Mol subpopulation are the characteristic changes in HLE-PSO patients, which might be related with the hyperlipidemia caused immune injury in patients. QHJ herbs could effectively improve the disproportion of Mo2a subpopulation.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effect of specific blockage of mutant p53 gene by individualized antisense RNA in vitro.</p><p><b>METHODS</b>Mutation status of p53 in human breast cancer cell lines was determined by immunocytochemical staining, PCR-SSCP and sequencing. Single strand antisense transcription system targeting specific p53 mutation site (mt-p53) was constructed, and corresponding antisense RNA was prepared. The hybridization of antisense RNA with its corresponding mt-p53 gene was confirmed by in-situ hybridization. Human breast cancer cells were transfected with antisense RNA by cationic liposome-mediated method. Time course of effects of antisense RNA was investigated by immunocytochemical staining and cell growth inhibiting assay. Expression of mt-p53 protein was examined by Western blot. Cell proliferation was evaluated by MTT assay and cell cycle distribution was determined by flow cytometry (FCM). Apoptosis was determined by TUNEL assay.</p><p><b>RESULTS</b>Mutation of p53 exon 8 was found in MDA-MB-231 cells and antisense transcription system (pGEM3zf (+/-) p53exon8) was then constructed successfully. In transfected MDA-MB-231 cells, hybridization signals were observed in cytoplasm. Fourth-eight hours after transfection, the antisense RNA (ASp53exon8'RNA) had a significant retarding effect on p53 related proliferation inhibition, along with a decrease of p53 protein expression.</p><p><b>CONCLUSIONS</b>ASp53exon8'RNA specifically blocks mt-p53 gene expression, resulting in an inhibition of MDA-MB-231 cell proliferation. Such an approach may be used as a therapeutic option against human malignancy.</p>

ABSTRACT

Objective To investigate the effects of heparin coating on intimal hyperplasia in implanted decellularized xenografts.Methods The resected canie carotid arteries were decellularized,and heparin coating was partially performed.Eighteen rabbits were divided into non-heparin-coated group(n=9)and heparin-coated group(n=9).During implantation,only the left carotid between the anastomotic stoma was ligated.Doppler ultrasonography was performed 1,3 and 12 weeks post-implantation to measure the luminal diameter,and the hemodynamic parameters such as PSV,RI and PI were calculated.All animals were sacrificed,histological observations were conducted at 12 weeks post-implantation,and I/(I+ M)was calculated.Results Except for 1 week post-implantation in the ligated side,the luminal diameters in non-heparin -coated group were significantly smaller than that of pre-implantation.Besides,those of the non-ligated side at each time points were significantly smaller than the ligated side(P

ABSTRACT

<p><b>OBJECTIVE</b>To observe the clinical efficacy of Qi-supplementing and blood-activating (QSBA) in treating diabetic peripheral nephropathy (DPN).</p><p><b>METHODS</b>Sixty-eight type 2 diabetes mellitus patients with Qi deficiency-blood stasis Syndrome were randomly divided into two groups, the neurotrophic agents were used in both groups, while QSBA herbs were used in the treated group additionally. The treatment course was 2 months. Blood glucose (BG), triglyceride (TG), total cholesterol (TC) and nerve conduction velocity (NCV) were detected before and after treatment.</p><p><b>RESULTS</b>After treatment, the BG, blood lipid, NCV were improved significantly (P < 0.05) in both groups, but the improvement was more significant in the QSBA treated group than in the control group (P < 0.05).</p><p><b>CONCLUSION</b>QSBA, in treating DPN, can not only improve its symptoms, but also ameliorate the NCV.</p>
